Calmodulin-binding proteins as calpain substrates.

چکیده
Calcium-dependent neutral protease, also called calpain, has attracted increased interest in recent years. Its properties include an absolute dependence on Ca2l for activity (micromolar and millimolar concentrations of Ca2l for calpain I and calpain II, respectively), and a pH optimum between 7 and 8. It is isolated from the cytosolic fraction of various animal tissues or cells but it may be translocated to the cell membrane under certain conditions (for previous reviews see Murachi, 1983a; Zimmer-

Neuronal nitric oxide synthase and calmodulin-dependent protein kinase IIalpha undergo neurotoxin-induced proteolysis.
Calpain (calcium-activated neutral protease) has been implicated as playing a role of neuronal injury in cerebral ischemia and excitotoxicity. Here we report that, in addition to extreme excitotoxic conditions [N-methyl-D-aspartate (NMDA), alpha-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA), and kainate challenges], other neurotoxins such as maitotoxin, A23187, and okadaic acid also...
متن کاملCalmodulin interacts with ATP binding cassette transporter A1 to protect from calpain-mediated degradation and upregulates high-density lipoprotein generation.
OBJECTIVE To investigate the interaction of ATP-binding cassette transporter A1 (ABCA1) with calmodulin in relation to its calpain-mediated degradation because many calpain substrates bind calmodulin to regulate cellular functions. METHODS AND RESULTS The activity of ABCA1 is regulated through proteolysis by calpain. An immunoprecipitation and glutathione S-transferase pull-down assay reveale...
متن کاملUsing Calmodulin-Affinity Capture to Study the Rat Brain Calmodulin Binding Proteome and its Vulnerability to Calpain and Caspase Proteolysis
Calcium dyshomeostasis is involved in neuropathological conditions such as traumatic brain injury (TBI), stroke and neurodegenerative diseases. Under such conditions in the brain, calmodulin binding proteins (CaMBPs) are particularly vulnerable to two abnormal processes: (1) over-activation by calcium bound calmodulin (CaM), and (2) proteolytic processing by calpains and caspases. In this study...
متن کاملCalpain I activation is not correlated with aggregation in human platelets.
Calpain-catalysed hydrolysis of platelet substrates such as cytoskeletal and calmodulin-binding proteins, and of protein kinase C, is assumed to contribute to platelet aggregation. We have measured calpain I activation by immunoblotting, and [Ca2+]i (cytoplasmic Ca2+ concn.) by fura-2 fluorescence, in parallel with measurement of aggregation, in stirred human platelets treated at different [Ca2...
متن کاملAmino-terminal conserved region in proteinase inhibitor domain of calpastatin potentiates its calpain inhibitory activity by interacting with calmodulin-like domain of the proteinase.
Calpastatin is a widely distributed endogenous inhibitor protein specifically acting on calpain (Ca(2+)-dependent proteinase) and is known to interact with the calmodulin-like domain (CaMLD) of the proteinase in a Ca(2+)-dependent fashion. The calpastatin molecule consists of four inhibitory domains (domains 1-4) with mutually homologous sequences in three regions designated as A, B, and C. Aci...
